Immunoblotting is a widely-used and powerful technique for the detection and identification of proteins using antibodies. The process involves the separation of sample proteins by polyacrylamide gel electrophoresis followed by the transfer of the separated proteins from the gel onto a thin support membrane. The membrane binds and immobilizes the proteins in the same pattern as in the original gel. The membrane (or “blot”) is then exposed to a solution containing antibodies that recognize and bind to the specific protein of interest. The antibodies bound to the membrane are detected by any of a variety of techniques, usually involving treatment with a secondary antibody.
